FRANÇAIS : Trois groupes fonctionnels de la molécule de delta 9-THC sont impliqués dans l'interaction ligand-récepteur: le C11, l'hydroxyl phénolique et la chaîne latérale. L'utilisation d'une série d'analogues substitués en C11, possédant une chaîne latérale diméthylheptyl, permet d'augmenter considérablement l'affinité pour le récepteur et le potentiel pharmacologique. Les effets comportementaux sont étudiés chez la souris. Les résultats permettent d'affirmer que les trois groupes fonctionnels n'interagissent pas de la même façon avec les récepteurs, que le C11 n'est pas la position essentielle et que des substituants en C9 pourraient réagir avec le même site mais différemment. ENGLISH : The basic premise underlying the cannabinoid pharmacophore is that at least three functional groups are involved in the interaction between the ligand and the receptor: C11, the phenolic hydroxyl and the side chain. In order to assess the importance of the C11 position and the side chain, a series of C11 substituted analogs were prepared which contained a dimethylheptyl side chain. Incorporation of a dimethylheptyl side chain enhances both pharmacological potency in mice and receptor affinity. Results show the improbability that all three of the functionnal groups are interacting in a similar fashion with the receptor and that the C11 position is not an essential requirement for activity. It is also possible that substituents in the C9 region are interacting somewhere within or near the same site, but differently. (From authors' abstract) |
