FRANÇAIS : La métyrapone est reconnue pour atténuer les effets comportementaux de la cocaïne chez le rat. Les effets de la métyrapone sont étudiés sur un autre psychostimulant, la d-amphétamine. Une pré-administration de métyrapone (3X100mg/kg), contrairement à l'hypothèse de l'étude, provoque une augmentation nette de l'activité locomotrice et des effets induits par la d-amphétamine. Le fait que le trisolane, un autre inhibiteur de la synthèse des corticostéroïdes, ne modifie pas les effets comportementaux de la d-amphétamine suggère que la propriété d'inhiber la synthèse des corticoïdes ne constitue pas le mécanisme sous-jacent de l'action de la metyrapone. Son mécanisme d'action pourrait être lié à l'inhibition de la débrisoquine hydroxylase (cytochrome p450 2D1), une enzyme impliquée dans le métabolisme de la d-amphétamine. ENGLISH : The clinically utilized corticosteroid synthesis inhibitor metyrapone attenuates the behavioural effects of cocaine in the rat. Given the potential therapeutic implications of this interaction, we felt it important to determine if metyrapone's action would generalize to another widely abused psychostimulant, namely d-amphetamine. However, rather than producing attenuation, metyrapone preadministration (3 x 100mg/kg) markedly enhanced both the locomotor activating and stereotypy-inducing actions of d-amphetamine (dose equivalent to 2.5 mg/kg free base). The fact that the corticosteroid synthesis inhibitor trilostane did not affect the behavioural action of d-amphetamine suggests that inhibition of corticoid synthesis does not underlie the action of metyrapone. Instead, it is argued that inhibition of debrisoquine hydroxylase (cytochrome p450 2D1), an enzyme involved in the metabolism of d-amphetamine, represents the critical mechanism of action.
