Dans toute société humaine, des individus ont tendance à utiliser des substances capables de modifier leurs sensations. L'application des méthodes expérimentales et des techniques actuelles en neurosciences et en clinique permet de comprendre le pourquoi biologique de cette tendance. Elle permet aussi de saisir les étapes conduisant du plaisir à la pathologie et à la dépendance. Ces connaissances amènent à pratiquer des traitements mieux ajustés et plus efficaces. (A partir du résumé d'auteur.) ENGLISH: In her lectures presented in Geneva and Lausanne, in September 2000, Prof. M.J. Kreek outlined the roles played by the genome and by the neurobiological disorders induced by the use of opiates, cocaïne, ethyl-alcohol and other drugs. From this study, it appears that pharmacodependence is a brain disease. Thus, Kreek's group has shown that in drug addicts, the u receptor may be changed by alleles of an inherited gene responsible for this cell receptor. Long-lasting disorders in the brain circuitry of "reward" follow drug use, while developmental and environmental conditions also intervene in the prognosis. Thus, it is known that opiates modify the normal function of the hypothalamo- pituitary-adrenal axis implied in stress reactions. Therefore drug addicts must be seen as patients and not as criminals and their treatment must include correction of the induced biological disorders. For opiate addicts, methadone was shown by Prof. Kreek to act in this way. (Author's abstract.) |
